Dr. Pam Mellon, Core Leader

Dr. Pamela Mellon has developed the UCSD transgenic mouse facility and will oversee a highly productive core aimed at developing genetically altered mice. The study of genetically manipulated mice is an essential feature of the UC San Diego SBRP. This core will take advantage of current and new technologies to translate genetic information into genetic experiments in the mouse. As environmentally activated target genes or other known genes of interest are identified, these genes will be studied further by genetic manipulation of function in transgenic mice. The core will assist in the design of transgenic and knock-out mice and will create genetically modified mice using injection of transgenes, the transfection of embryonic stem cells for knock-out studies and injection into blastocysts. The core is highly successful and has an excellent track record in assisting in the characterization, support and maintenance of the mouse colonies in conjunction with the Superfund investigators. The Core is also available to preserve mouse lines through cryogenic embryo preservation, to assist with mice breeding problems through ovary transplant or in vitro fertilization, and to provide breeding pairs of some lines maintained through the core. In addition, the core is establishing methods for the creation of transgenic mice using Bacterial Articficial Chromosomes (BAC) as well as Recombinant Lentivirus.
